Targeting extracellular vesicle delivery to the lungs by microgel encapsulation

چکیده
Extracellular vesicles (EVs) secreted by stem and progenitor cells have significant potential as cell-free ‘cellular’ therapeutics. Yet, small EVs (<200 nm) are rapidly cleared after systemic administration, mainly the liver, presenting challenges targeting to a specific organ or tissue. Microencapsulation using natural nano-porous hydrogels (microgels) has been shown enhance engraftment increase survival of transplanted cells. We sought encapsulate within microgels target their delivery lung virtue size-based retention pulmonary microcirculation. Mesenchymal stromal cell (MSC) derived were labelled with lipophilic dye (DiR) encapsulated agarose-gelatin microgels. Endothelial bone marrow macrophages able take up in vitro, but less efficiently than uptake free EVs. Following intrajugular microgel selectively retained lungs for 72h, while liver. Furthermore, microgel-loaded demonstrated greater cells, particular CD45+ immune assessed flow cytometry compared may be novel tool enhancing targeted future therapeutic applications.

Targeting Drug Delivery to the Lungs by Inhalation
Most drugs targeted to the respiratory tract are used for their local action. For example, ephidrine for nasal decongestion, beta-2 agonists for bronchodilatation, and inhaled steroids to suppress the inflammation seen in asthmatic airways. Since the drug is delivered directly to its required site, only a small quantity is needed for an adequate therapeutic response, and consequently there is a...
متن کاملThe Obstacles to Current Extracellular Vesicle-Mediated Drug Delivery Research
Extracellular Vesicles (EVs) are nanometer-sized cell-derived membrane vesicles that are released by donor cells and play an important role in intercellular communication. In this short communication, we discuss the obstacles currently faced in EV-mediated drug delivery research. The commonly used vehicle for drug delivery in prevalent practice are liposome's which are synthetic vesicles, these...
متن کاملDrug delivery to the Lungs 21.
Drug Delivery to the Lungs 21 was focused exclusively on delivery technologies of medicines for the treatment of diseases that are 'local' to the respiratory tract or for wider 'systemic' distribution. Therefore, the range of diseases that can be treated via delivering drugs to the lungs is large and diverse. This diversity means that the delivery technologies (device and/or formulation) are al...
متن کاملUnilamellar vesicle formation and encapsulation by microfluidic jetting.
Compartmentalization of biomolecules within lipid membranes is a fundamental requirement of living systems and an essential feature of many pharmaceutical therapies. However, applications of membrane-enclosed solutions of proteins, DNA, and other biologically active compounds have been limited by the difficulty of forming unilamellar vesicles with controlled contents in a repeatable manner. Her...
متن کاملExtracellular vesicle in vivo biodistribution is determined by cell source, route of administration and targeting
Extracellular vesicles (EVs) have emerged as important mediators of intercellular communication in a diverse range of biological processes. For future therapeutic applications and for EV biology research in general, understanding the in vivo fate of EVs is of utmost importance. Here we studied biodistribution of EVs in mice after systemic delivery. EVs were isolated from 3 different mouse cell ...
